We have a 2000 Land Rover discovery and in the snow we noticed that the left rear wheel wasn't moving. We lifted the truck in the air and we could spin the wheel but it doesn't move when the engine is running. What could be wrong with this?

Probably nothing. The wheel with the least resistance will spin when there is no load. When it's in the air, grab the wheel that's spinning and hold it, the opposite wheel will then start to spin.

is it supposed to have sure grip diffs?

no landrovers were ever fitted with sure grips from the factory, so the wheel will turn. the way they stop wheelspin is by using the brakes to stop the spinning wheel via a sensor just like abs, so the sensor is probably sensing wheel movement whem the engine is on and stopping it via the caliper
