What does it take to change the rear main seal with the motor in the car? Does it require the trans to come down?

depends... if is the rubber seal, just need to drop the pan, windage tray and seal cap. Then push on one side to pull it out from the other side... then insrt the new one reversing operation.

if is the rope seal, need to remove crank, however I made to fit just loosening the crank ( and flywheel from troque converter of course ) and drop it a little bit without remove main caps and then fit the rope seal :D

on any case, tranny keeps in place. You don't get access to that area trhought tranny side
